“Purity” by LCMS (the standard test used by all the consumer peptide testing labs) means most of the measured signal comes from one main component with the expected mass after the sample has been passed through a 0.22 µm filter, which removes particulates before analysis. That filtering step means the test only looks at the filtered liquid, not what you’re getting out of the vial and injecting.
Larger impurities like aggregates or particulate contaminants are taken out beforehand and won’t show up in the result.
“Purity” in this case means the filtered liquid matches the molecular weight of what’s expected compared to a reference sample of rHGH, but it doesn’t prove everything is correct. It can miss subtle chemical changes (like certain oxidations), and structural issues such as improper folding (meaning the molecule may be rHGH but it won’t “dock” properly with the receptor). So a “high purity” result really indicates the filtered sample is mostly one species with the right mass, not that the vial contents are free of impurities or fully correct in structure or function. Thats why a peptide can be “99% pure” but still fail a sterility test, since bacteria are too large to pass through the filter.
Since no single device can do it all, pharmaceutical labs use (and the FDA requires) multiple methods, a technique called “orthogonal testing” to overcome the limitations of LCMS (e.g., peptide mapping with MS-MS, proper structure and folding using NMR, testing bioactivity to see how potent it is via test kits with special cells that have human growth hormone receptors, etc) to fully verify quality.
